Effect of Albumin Concentration and Preincubation Time on Motility and Acrosome Reaction in Bovine Semen

Abstract
This study was performed to investigate the effect of albumin and preincubation on acrosome reaction and motility of sperm. Frozed bull semen was thawed and incubated in Ham's F-10 media containing 0, 0.3, 3, 10 percent of bovine serum
albumin(BSA)
resperctively. The analysis of sperm motility lknad making of slides for observation of acrosome reaction was done in 2, 6, or 24 hour incubation.
@ES The results were as follows:
@EN 1. The incidence of ture acrosome reaction (TAR) in sperms which were incubated in a albumin free medium was highest to be 22 percent in 2 percent in 2 hour incubation.
2. The incidenece of TAR in sperms which were incubated in a medium containing 0.3 percent of BSA was highest to be 26 percent in 6 hour incubation, in a medium containing 3 percent of BSA was 35 percent in 2 hour incubation, and in a medium
containing
10 percent of BSA was highest to be 20 percent in 2 hour incubation.
3. In a albumin free medium, motility index was sharply decreased from 241.2 to 3.3 according to the length of incubation.
4. In a medium containing 0.3 or 3 percent of BSA motility index was sustained relatively well until 6 hour incubation, but was sharply decreased to 11.8 or 25.4 in 24 hour incubation, but in a medium containing 10 percent of BSA motility index
was
blunitly decreased to be 117.9 even in 24 hour incubation.
5. Sperm recovery rate from swim up preparation was as low as the highest value was at most 13.3 percent.
6. There was positive correlation between motility index and the incidence of TAR(r=0.6257.
In this study it was concluded that albumin was required for incubation of acrosome reaction in indirect manner. And as the incidnece of acrosome reaction itself was increased, motility rate of sperms was decreased according to the length of
incubation.
And, sperm recovery rate from swim up preparation was so low that thawed sperms would be used without swim up preparation in case of artificial insemination.
